Don’t stop at Native ChIP, complete the picture with Mass Spectrometry

We are very pleased to announce that we can now add mass spectrometry to our range of downstream applications. Our Chromatrap® Native ChIP kit is now fully compatible for mass spectrometry (MS) as a downstream process. Combining the solid state technology with MS enables researchers to obtain high quality datasets for chromatin proteomic profiling and mapping of histone modification patterns.

 

Technique Background

Chromatin immunoprecipitation (ChIP) is an immunoprecipitation technique commonly used for mapping the DNA-protein interactions in cells which are crucial for gene regulation.

The technique enriches DNA fragments to which a specific protein of interest or histone modification is bound. Enriched sites can then be identified by qPCR, hybridisation to a microarray (ChIP-on-chip), or by sequencing.

 

In our post-genomic era, mass spectrometry has become an invaluable versatile tool in the analysis of biomolecules. As a popular method of choice by researchers, MS is widely used to identify chromatin binding proteins and elucidate histone modification patterns.

 

In native ChIP (N-ChIP), the naturally occurring DNA-protein complexes are preserved by removing the need to fix cells prior to extraction. Although there are a number of published protocols outlining methods to performing N-ChIP, Chromatrap uniquely offers researchers an efficient kit with an optimized protocol specific for the technique.
