Human Control qPCR Primer Sets were designed to serve as positive or negative ChIP controls when performing chromatin immunoprecipitation with human samples. Each primer set is a mixture of forward and reverse primers that have been validated for qPCR of ChIP samples from multiple human cell lines.
In addition Chromatrap® supply forward and reverse primers on their own for common targets.
Chromatrap®’s Negative Primer Set 1 recognises the hypomethylated protein-coding gene UBE2B, this will not be picked up by methylation study assays. A 177 base pair fragment is amplified by this primer set. The enzyme that this gene encodes is required for DNA damage repair after replication has occurred.
It is a negative gene target for 5-methylcytosine.
Chromatrap®’s Negative Primer Set 2 recognises the promoter of the GAPDH gene. A 69 base pair fragment is amplified with Negative Primer Set 2.
It is a negative gene target for histone modifications that are associated with repressive transcription. This includes the following: H3K27me3, H3K9me3 and the transcription factor Sin3a.
Chromatrap®’s Negative Primer Set 3 recognises the promoter of the human B-Globin gene. A 121 base pair fragment is amplified using this primer set.
It is a negative gene target for active genes including H3K4me3 and RNA Poll II that are involved in initiating transcription, as well the Androgen Receptor (AR) gene.
Chromatrap®’s Negative Primer Set 4 recognises the promoter region of the protein-coding SAT2 gene. A 72 base pair fragment is amplified by this primer set.
It is a negative gene target for the acetylated histone mark H3K14ac, associated with active transcription.
Chromatrap®’s Negative Primer Set 5 recognises a region proximal to the promoter of Myt1, a zinc finger DNA-binding protein that is associated with repressive transcription.
It is a negative gene target for the acetylated histone mark H3K27ac.
