Background:

5mC (5-Methylcytosine) is a region of DNA that has been epigenetically modified through the addition of a methyl group at the 5th carbon position of cytosine. DNA methylation is a repressive histone modification that is associated with switching off gene expression. This methylation generally occurs at CpG islands and is also associated with genomic imprinting. Aberrant DNA methylation is linked to disease including many cancers.

A mouse IgG is included in this Antibody Primer Set as a negative control for the MeDIP experiment.

Background:

RNA polymerase II catalyses the transcription of DNA into mRNA and other small nuclear RNAs in eukaryotes. It contains a CTD consisting of conserved heptapeptide repeats. Phosphorylation occurs at serine and therine residues located in the CTD repeats to activate the RNA pol II. RNA pol II binds to several transcription factors in order to initiate transcription and therefore serves as an abundant antibody target for ChIP.

A mouse IgG is included in this Antibody Primer Set as a negative control for the ChIP experiment.

Background:

Histone 3 tri methyl K9 (H3K9me3) is a histone mark associated with repressed chromatin. It condenses and compacts the chromatin, restricting the transcription machinery from binding and carrying out gene expression. It is a marker of constitutive heterochromatin, and generally associated with gene silencing. As a histone mark, it is widely found throughout human cell lines, serving as an abundant antibody target for ChIP.

Background:

The androgen receptor (AR) is a steroid hormone receptor involved in the signalling of testosterone. It is a regulatory gene that controls gene expression through binding to androgen receptor elements (AREs). The AR is associated with both normal prostate and cancerous prostate cell growth at abnormal levels of the gene (Lamb et al, 2014, BJUI). Androgens are also heavily involved in the pathogenesis of endometrial cancer (D.A. Gibson et al, 2014, Endocr Relat Cancer). DHT (dihydrotestosterone) is a natural androgen hormone that can activate the androgen receptor (Mostaghel, 2014, IJBS).

Background:

Sin3a (Switch independent 3 homologue) is a transcriptional repressor and protein-encoding gene. It interacts with co-repressors and DNA-binding proteins such as REST, MXl1 and MXD1-MAX that allow Sin3a to bind to DNA and repress transcriptional activity. It acts interdependently with OGT (O-linked N-acetylglucosamine) to repress transcription alongside with histone deacetylation.
